Colorimetric Determination of Tyrosine BY G. CERIOTTI AND L. SPANDRIO

چکیده

The procedure described permits quantitative application of the Sakaguchi reaction. The greatest advantage of the method is the stability of the coloured product in butanol. The sensitivity, already high in the original method, is further increased, since the volume of the final coloured solution is reduced by the extraction. For concentrations of arginine ranging between 0-5 and 6 ,ug./ml. there is a linear relationship between quantity and colour yield and there is good reproducibility. The variability of the older method introduced by variability in the quality of urea, by nitrogen-bubble formation, by condensation on cuvettes and by rapid decrease of colour intensity is avoided. These factors are so important that each constitutes a serious obstacle to the quantitative application of the older methods. The empirical variations of hypobromite concentrations from one test to the other, suggested by Sakaguchi (1948), are also avoided. This is particularly important, as variations in hypobromite concentrations have a considerable effect on the colour yield. The a-naphthol-diacetyl reaction in the presence ofpropanol, performed by the method of Rosenberg et al. (1956), is undoubtedly an improvement over the original Sakaguchi (1925a, b; 1948, 1950, 1951) method, but it is less sensitive than the present method and less reproducible. In the present method the colour develops immediately, and then shows no significant change for a long time; the influence of temperature is also limited.

Improved direct specific determination of serum iron and total iron-binding capacity.

Serum iron is released from transferrin and reduced at pH 1.7 by treating serum with a 10 g/L ascorbic acid solution in 0.1 mol/L HCl. When ferrozine is added to this reagent, it forms a complex with iron that is as intensely colored as at higher pH values, and under these conditions no turbidity is produced. The second major interference, that from copper, is eliminated by adding 1 g of thiose...

Determination of nucleic acids in animal tissues.

As reported in a previous paper (l), a new microchemical method for the determination of deoxyribonucleic acid (DNA) has been devised which permits the evaluation of DNA in amounts as small as 2.5 y per ml. The method is particularly useful with very small quantities of tissue. For this reason a study was undertaken to learn the best conditions for the application of the method to the determina...

Analytical Performances of an Enzymatic Assay for the Measurement of Glycated Albumin

Background: Short to intermediate integrated glycemic control is best determined by glycated albumin (GA). This assay is appropriate when interpretation of glycated hemoglobin (HbA1c) is critical because of hemoglobinopathies, severe anemias, or other factors that affect red blood lifespan as hemodialysis. We evaluated a new assay based on the enzymatic quantification of GA by ketoamine oxidase...

Urea, creatinine, and glucose determined in plasma and whole blood by a differential pH technique.

We report the conditions (buffer composition and enzyme activity) required for estimating three frequently determined analytes--urea, glucose, and creatinine--by use of an improved version of the differential pH apparatus previously described (Clin Chem 29: 80-85, 1983). For each analyte, we used only one specific enzyme, thus avoiding a chain of auxiliary and indicator reactions. The method re...

Measurement of lipase activity by a differential pH technique.

This is a new electrochemical method for determination of lipase activity in biological fluids, including serum, plasma, and duodenal juice. Advantages of turbidimetric methods--short reaction time, and small sample and reagent volumes--are combined with those of titrimetric methods: measurement of absolute activity (i.e., no standardization required), saturated substrate conditions, and direct...
